I must say, this particular Barge run was very impressive- No NPC knocks and this exact route by nature there is zero things in the way like Asteroids etc.
I welcome you to test this exact example given.
Not sure how you fly your Barge, but mine, which is the same as what everyone else has, takes:
150 seconds to get to cruise.
About another 25 seconds to achieve 99 speed.
The Barge travels 6k every minute, in cruise.
The journey is approx. 56.3k from point A to B (no obstacles in the way).
Time at start before undocking was: 21:41:19
Time at docking was: 21:53:08
Journey time was: 11:49

Return journey was commenced as soon as I docked. Appeared from NPC Base facing the wrong way. Barge has a large turning circle.
So, say it took me 10 seconds to dock and undock.

Return journey time: 12:03
Total time: 23:52 (1,432 seconds) for a journey of 112.6k with a docking at mid point.

5ker was 4.05 times faster (granted 5ker undocked on the return journey facing the right way).
Now using the TL that is 9k from the start/finish point and goes right to the midpoint of the journey:

5ker was 8.23 times faster than the Barge and that is even with 18k of travel in cruise and using 1 Trade Lane there and 1 back.
Conclusion:
In a straight race, in open space, the 5ker is approx 4 times faster than a Barge, mainly due to the time it takes to get to top cruise speed for the Barge.
Using TLs the 5ker has a huge advantage. It takes a Barge about 30 minutes to go from Planet New London to the Leeds Jump Gate (just over 150k as the crow flies). I didn't time the journey for a 5ker using the TLs, but I don't think it takes anywhere near 5 minutes.
When it comes to any System with asteroids/ice clouds/debris etc, and there are quite a few, this is where the Barge greatly suffers. A 5ker can easily pick its way through the obstacles, whereas the Barge easily gets stuck and this greatly slows down the Barge especially as it takes it about 25 seconds to get from 0 speed to 99 if it is already in cruise.
