If you are running windows (which I sure you are), You have a TEMP folder on your computer. Running programs, and auto-extracting installers and other kinds of programs will place files in there while they are running. Its basically a cache. Usually when a program is shut down, the files it used are deleted. Sometimes however, they aren't. Sounds like something used a LOT of space, and it somehow 'left the building'. You can manually empty it using "Disk Cleanup" or whatever its called.
EDIT: Do you have "Hibernation" avaible on your computer?

Another thing to watch for is how much you've allocated to virtual memory. That can eat your HD alive.

That happened to me too.. Well I deleted some movies from my hard drive... and then checked its size and it was around 20 gb free space...
I turned off computer and went to bed...
Next day I was working on some text files (couple kb) and after editing couple files I deleted rest ... Then i emptied my Recycle bin and had 45 gb of free space...
Maybe you did similar thing.... Files that aren't directly deleted stay in cache on hard drive... (at least i think so)
